چکیده

The nucleotide sequence of rainbow trout (Salmo gairdneri) 5.8 S rRNA was determined to be p(C)-A-A-C-U-C-UIO LO SO Um-A-G-C-G-G-U(m)-G-G-A-U-C-A-C-U-C-G-G-C-U-C-G-U411 51, G-C-G-U-C-G-A-U-G-A-A-G-A-A-C-G-C-A-G-C-G-C-U-A-G80 ill C-(~V)-G-C-G-A-G-A-A-C-U-A-A-U-G-JI-G-A-A-U-U-Gm-C-AX0 911 1,N, G-G-A-C-A-C-A-U-U-G-A-U-C-A-U-U-G-A-C-A-C-U-U-C-G1 LO lL,l A-A-C-G-C-A-C-U-U-U-G-C-G-G-C-C-C-C-A-G-G-G-G-U-U,:,,I ,111 150 C-C-U-C-C-U-G-G-G-G-C-U-A-C-G-C-C-U-G-U-C-U-G-A-G,w U-G-U-C-G-C-U-U. The major molecular species is 161 nucleotides long; the sequence differs in only eight nucleotides from that of Man. An estimate of the secondary structure retains essentially all of the features previously described in mammalian 5.8 S RNAs with several interesting variations. The very stable G-C-rich hairpin loop is extended by two additional hydrogen bond pairs; the A-U-rich loop is shorter; and a third 2’@-methylation occurs at residue 8 close to the under methylated U(m) residue at position 14. Comparisons with other known sequences indicate that in eukaryotes 5.8 S rRNA is more highly conserved than 5 S RNA species.

Cloning and sequencing of rainbow trout (Oncorhynchus mykiss) interferon regulatory factor 7

Interferon regulatory factor 7 (IRF7) gene was cloned from a subtractive cDNA library constructed with mRNAs obtained from rainbow trout (Oncorhynchus mykiss) macrophage cell line (RTS-11). Using expressed sequence tag clones of submitted IRF7 amino acid sequences, specific primers were designed. Results showed that IRF7 cDNA contains an ORF of 1251 nucleotides that translates into a 416 resi...

Structural analyses of mammalian ribosomal ribonucleic acid and its precursors. Nucleotide sequence of ribosomal 5.8 S ribonucleic acid.

The nucleotide sequence of ribosomal 5.8 S RNA (also known as 7 S or 5.5 S rRNA) from Novikoff hepatoma ascites cells has been determined to be (see article). Estimations of the secondary structure based upon maximized base pairing and the fragments of partial ribonuclease digestion indicate that there may be five base-paired regions in the molecule, three forming a folding of the termini and t...

Characterization of a complete immunoglobulin heavy-chain variable region germ-line gene of rainbow trout.

A germ-line heavy-chain variable region (VH) gene (RTVH431) has been isolated from a rainbow trout (Salmo gairdneri) and characterized by complete nucleotide sequencing. It is characteristic of VH, as shown by the conserved octamer and TATA motif in the 5' region, the heptamernonamer recombination signal sequence in the 3' region, and the 18-amino-acid-long hydrophobic leader interrupted by an ...

Purification of a major serum protein of rainbow trout (Salmo gairdneri) homologous to the third component of mammalian complement.

The present paper describes the purification and characterization of a major serum protein of the rainbow trout (Salmo gairdneri) required for complement-related functions (target cell lysis and opsonization). This protein, termed C3-1, was identified as the third component (C3) of rainbow trout complement. It is homologous to component C3 of human and other mammalian complements based on its s...
